ProSentium™ - the T cell epiproteome at your fingertips

ProSentium™ is ProImmune's proprietary database of naturally occurring self-peptides presented by MHC to T cells, as determined by tandem mass spectrometry.

The data in the ProSentium™ database have been obtained with our unique ProPresent® antigen presentation assay and is now available for licensing.

In 2011, ProImmune introduced the ProPresent® antigen presentation assay, a method that directly identifies peptides presented by MHC molecules on the surface of antigen presenting cells through sequencing mass spectrometry.

 


 

Over recent years we have significantly improved our methodology making it possible for ProPresent® to identify tens of thousands of self-peptides from each donor sample. When the technique is run over dozens of donor samples this reveals a unique profile of proteins and peptide antigens that are visible to the immune system, including the prevalence with which these sequences are observed. This information is essentially a snapshot of the T cell epiproteome and it covers a vast range of antigens with high disease relevance.

How ProSentium™ Works

By request we can investigate and report the peptide hits from our ProSentium™ collection based on pre-existing assay data for a specific protein or protein family. Results will be reported as overlapping nested peptide sets, similar to the tabular reports for the ProPresent® assay. Reported data are available for exclusive or non-exclusive license.

How do you know what you will get?

We can generate free of charge preview reports for your protein of interest which clearly set out the number of donors available for reporting, their tissue type distribution and the number of peptide hits and unique peptide hits that can be found at a minimum statistical significance. Some proteins show many presented sequence and some show only a few key presented sequences. Each case can be very informative in making drug development decisions.

What kinds of protein are in the ProSentium™ collection?

The proteins found in ProSentium™ span many major groups of high-value disease targets including in the following mutually non-exclusive groups:

  • Tumour associated antigens

  • Cell surface receptors of all types

  • Cytoskeletal proteins

  • Intracellular enzymes

  • Immunologically relevant proteins including Ig receptors, cytokines, complement related proteins

  • HLA molecules

  • Lipid transport proteins

  • Major groups of serum proteins

  • GPCRs

  • Proteins associated with cancer, autoimmune diseases, cardiovascular disease, bleeding disorders, neurodegenerative disease, transplantation antigens and many more

What ProSentium™ information can be used for:

ProSentium™ data can be used to investigate immune responses against such classes of proteins, how they are associated with specific HLA tissue types and guide how interventions to reengineer such proteins could lead to new beneficial therapeutic approaches. Prosentium™ is effectively very similar to our ProPresent® antigen presentation assay service, except that no further experimental work has to be done. The data is ready for analysis and reporting and will be available from many more donor samples than could reasonably run for an individual ProPresent®  project.

As such ProSentium™ data can also be readily combined with further ProPresent® and other in vitro assay services such as ProImmune REVEAL MHC peptide binding assays, T cell and DC T cell proliferation assays and MHC multimer technology available at ProImmune. As part of our overall assay suite ProSentium™ can therefore support pivotal decisions with physical evidence in game changing therapeutic approaches that target or interact with the immune system.

How does ProSentium™ compare with  Immune Epitope Database (IEDB) data?

ProSentium™ only focusses on sequences that are presented by antigen presenting cells (APCs) on MHC molecules and that are identified by LC-MS/MS. In the IEDB, a subset of sequences have been identified with similar methods. Our collection of presented self-peptides is significantly larger than that in the IEDB and also more diverse, unbiased against a specific project objective.

We currently offer ProSentium™ data on a bespoke basis only. Please contact us with your protein or protein family of interest and we will put together a custom licensing proposal.

  • Picture of presented cell-cycle and other oncogene protein targets.

  • Can inform drug engineering projects.

  • Development of tolerogenic drugs.

  • Identification of potentially immunogenic regions in therapeutic proteins.

  • Identify options for re-engineering, or deimmunizing proteins.

  • Can be combined as an information tool with more specific antigen presentation studies using the ProPresent® assays as well as ProImmune’s other epitope mapping and validation tools.
